For this week’s lab report, write out a step-by-step protocol for your Final Project (based on the hypothesis you selected and its associated data collection table). This will be easiest if your group works together on the answers. Make sure you create a protocol that includes all the steps (and materials) you will need to complete your project–including repetitions (for greater numbers), statistical analyses, and controls. The Effect of Light Color on Chlorophyll Production in Plants Objective To investigate how different colors of light affect chlorophyll production in plants. Materials – Potted plant specimens (e.g., bean plants) – Blue, red, and green LED lights – Dark box or room for control group – Spectrophotometer – Mortar and pestle – Solvent (e.g., ethanol) – Filter paper – Measuring equipment (beakers, graduated cylinders) – Notebook and pen for recording data Procedure 1. Chlorophyll Extraction: – Grind gathered leaves in a mortar and pestle with a tiny amount of solvent (e.g., acetone or ethanol). – Filter the solution to get a clear chlorophyll extract. 6. Spectrophotometric Analysis: – Use a spectrophotometer to measure the chlorophyll extract’s absorbance at wavelengths specific to chlorophyll a (about 664 nm) and chlorophyll b (around 647 nm). – Write down the absorbance readings for each sample. 7. Repetition: – Each repetition includes a new set of exposed leaves in bright color conditions. This means that we only use another set of leaves from the dark box and control group and repeat the abovementioned testing procedure. 8. Data Analysis: – Use suitable statistical tests (e.g., spectrophotometer) to compare chlorophyll absorbance between light exposure groups. – Determine whether there are any significant differences in chlorophyll production between the groups. 9.
