Normal cell growth and proliferation is governed by conserved signaling pathways, which when turned on can elicit specific growth patterns that ultimately expand the number of cells within a given tissue. However, when specific genes within these growth regulatory pathways are aberrantly expressed, dysregulated growth and proliferation ensue, which inadvertently results in disease.
Since, research has shown that there are many key proteins that operate either individually or in concert with other factors to regulate normal cell growth, your assignment is going to be about addressing different questions based on very specific circumstances, and to provide answers and detailed working plans for each of the following questions:
You have selected a gene and determined its genomic structure as well as its cDNA, for which you have generated a restriction map, and you have also identified potential miRNAs that might be involved in regulating its expression. Based on your initial observations, you determined that your gene of interest is overexpressed in a wide spectrum of solid and liquid tumors. To gain insight into its mechanism of action, you cloned the cDNA of your gene of interest into a mammalian expression vector, and you transfected it into an immortalized non-transformed human fibroblast cell line. Cell growth studies indicated that overexpression of the cDNA of your gene of interest results in hyperproliferation of human fibroblast cells, and siRNA-mediated knock down results in restoring normal cell growth.

To understand how the cDNA of your gene of interest influences cell growth, you conduct a
yeast two-hybrid assay and you discover that among its binding partners are cyclin-dependent kinase 1 (CDK1), CDK2 and CDK4/6. You immediately suspect that the protein produced by the cDNA of your gene of interest has an effect on kinases that control cell cycle progression.
b) Describe in detail how you would test the effect of the protein produced by the cDNA of your gene of interest on one of the CDKs, and include controls in
your experimental design. (2 points)
To shed more light on the mechanism by which expression of your protein of interest is increased in human solid and liquid tumor cells, you examine your gene of interest mRNA levels and discover that its levels are 10-fold lower compared to normal tissues. In addition, you also measure the levels of your protein interest and discover that it is 5 times higher in cancer cells compared to normal tissues.
C) Explain the lack of correlation between increased protein expression and decreased mRNA levels in tumor cells? (1.5 points)

You have measured the rate of transcription and half-life of you gene of interest mRNA and found that they are the same in normal and cancer cells.
You have also measured you gene of interest protein stability by conducting pulse-chase experiments in the presence of labeled cysteine and methionine, and discovered that protein stability is the same in normal and cancer cells.
d) What experiment do you propose to restore normal cell growth based on the explanation you have provided in C.1.? Please be as detailed as possible and
include controls in your experimental design. (1.5 points)
