Introduction:
The purpose of the DNA Collection Protocol is to provide proper instruction in the collection, packaging, documentation, storage, submission and disposition of animal DNA from alleged dogfighting dogs.
Collection:
1. A minimum of three buccal swabs will be obtained from each Pit bull type dog seized in a dogfighting investigation.
2. The forensic veterinarian examining the dog will collect the samples at the time of examination at the temporary shelter. There may be limited occasions were the samples will need to be obtained in the field.
3. Cytology brushes should be utilized to obtain 3 buccal swabs. a. Gloves must be worn by all who handle these brushes and prevent cross contamination. b. Remove the brush from the packaging and place the brush, one at a time, against the
inside of the cheek and swirl for ten seconds. c. The dog should not eat or drink at least 20 min prior to sampling. d. Following collection the brushes should be allowed to air dry and then place in an
unused double swab box together.
Packaging:
1. Swabs from multiple animals should NOT be packaged together. 2. The three cytology brushes should be placed together back in their original packaging, which
should then be placed in a properly labeled and sealed paper envelope. All packages must be properly identified. The container should be labeled before the item is placed into it, so there is no confusion once the container is sealed.
a. The container should state: i. Case number
ii. Item number iii. Agency iv. Location of collection (i.e. Temp Shelter) v. Brief description of the item it contains including where it was collected from
vi. Date and time the item was collected vii. Initials of the collecting veterinarian
b. It is not necessary to mark the samples individually, however the number of samples should be marked on the exterior of the container (Ex. Item 102A – (2) buccal swabs).
a. The bag or envelope must be completely sealed with evidence tape, or clear packing tape may be used to seal the container and a small piece of evidence tape used as a token seal. The evidence tape should be marked with the veterinarian’s initials and the date. The markings should cross over the evidence tape and carry onto the packaging.

Documentation:
1. Following collection of the buccal swabs and brushes the items of evidence should be logged in the animals individual Medical Evidence Log.
Storage:
1. The item(s) of evidence should be turned over to the evidence custodian at the end of each day. 2. Following the collection of all buccal swabs and brushes, these items are to be transferred (hand
delivered or shipped) to the ASPCA Veterinary Forensic Science Center for long term storage.
Submission:
1. The lead forensic veterinarian will determine if a sample is to be submitted for testing and will ensure that the necessary samples are submitted to the indicated testing facility in accordance with the Veterinary Forensic Services Evidence Submission Protocol.
Disposition:
1. Buccal swabs and brushes will be disposed of under the written direction of the primary agency and in accordance with the Veterinary Forensic Services Evidence Submission Protocol.
