Introduction
Cellobiase breaks down the 1,4 B-glucoside linkages in cellobiose is produced by many organisms. Fungi, such as molds, yeast, and mushrooms produce this enzyme and can secrete it to digest cellobiose to produce glucose for energy usage. Many bacteria also contain cellulolytic enzymes and cellobiase to break down plant cell walls. These bacteria can be found in the second stomach (rumen) of many hoofed animals such as cows and also in the gut of termites. This experiment will be performed over two lab periods. During the first period, you will evaluate the effects of concentrations of the enzyme and substrate on the reaction. During the second lab period, you will evaluate the effects of pH and temperature and examine the enzyme activity of cellobiase that you extract from a mushroom sample.NOTE: You will be using a variety of different enzyme and buffer solutions during this exercise. Be sure to use the correct solutions for each section of the exercise or you will not obtain usable data. DO NOT DISCARD ANY STOCK SOLUTIONS OR STANDARDS UNTIL THE ENTIRE EXERCISE HAS BEEN COMPLETED!
